Treatment of two pediatric patients with refractory systemic lupus erythematosus using CD19-targeted CAR T-cells.
This report describes two paediatric patients with refractory systemic lupus erythematosus treated with CD19-targeted CAR T-cells; both tolerated the therapy well and showed marked efficacy at five and four months respectively — early, two-patient evidence in a population where CAR T-cell therapy was previously unexplored.
Background
Chimeric antigen receptor (CAR) T-cell therapy has emerged as a promising strategy for autoimmune diseases, particularly systemic lupus erythematosus (SLE), but its use in paediatric SLE had not been explored.
Study design
Case report of two paediatric patients with refractory SLE who received an infusion of CD19-targeted CAR T-cells.
Key findings
Both patients tolerated CAR T-cell therapy well, and the treatment demonstrated remarkable efficacy after five months (patient 1) and four months (patient 2). With only two patients and short follow-up, these observations are hypothesis-generating and do not establish efficacy or long-term safety in children.
